The blockade lasted for at least four hours at the Chaman crossing in Pakistan's Balochistan province.
On Thursday Afghan border guards fired on Pakistani and Afghan tribesmen in the area who were holding a demonstration.
Local residents say that several people were wounded in the incident.
The protest was reportedly about a decision by Afghanistan's government to build a customs office at the border using private land. Many of the landowners in the area come from both sides of the border.
Friday's blockade was in protest against the shooting.
A Pakistani border guard spokesman told the Associated Press news agency that the blockade ended after security officials held talks with tribal elders and assured them their grievances would be passed on to the Afghan authorities.
On Thursday, more than 200 militants from Afghanistan stormed a Pakistani village in tribal areas near the border, wounding at least two women.
